HTS of Histidine-tagged Proteins

His MultiTrap FF and His MultiTrap HP are prepacked 96-well filter plates for fast screening and rapid, high-throughput parallel purification of histidine-tagged recombinant proteins. Reproducibility is excellent and purifications are directly scalable to larger columns. His MultiTrap FF and His MultiTrap HP, packed with Ni Sepharose 6 Fast Flow and Ni Sepharose High Performance respectively, are two new convenience products for purifying 96 samples with up to 1 mg of histidine-tagged protein/well in as little as 60 minutes. Users can load unclarified lysates directly, thereby decreasing sample preparation time, thus simpliying the screening workflow. Screening membrane and soluble proteins is a typical task.

The familiar 96-well plate format also allows great flexibility; His MultiTrap FF and His MultiTrap HP can be used manually using centrifugation or vacuum as well as in automated robotic systems. Consistent plate-to-plate and well-to-well performance ensures high reproducibility.Fast kinetics eliminate potential leakage problems caused by adding detergents to buffers and samples. Short purification times also minimize adverse effects such as degradation and oxidation of sensitive target proteins.Ni Sepharose has low nickel (Ni2+) leakage and is compatible with many additives used in protein purification. Availability in larger prepacked columns and as bulk media simplifies scale-up. The standard-dimension 96-well plate is made of bio-inert polypropylene and polyethylene. Each well is prepacked with 50 µl medium. Plates are supplied in packs of four.
