BD ACT:One is a highly-sensitive and cost effective tool for screening G-protein coupled receptors (GPCRs) that signal through Gs or Gi G-proteins. Using a natural biosensor approach, this technology has many advantages over biochemical or other live cell technologies. The proprietary biosensor is developed around a modified rat olfactory cyclic nucleotide gated (CNG) channel – a fairly non-discriminatory ion channel that responds to cAMP and cGMP. By engineering the CNG to enhance the cAMP binding affinity and reduce the cGMP binding affinity, Atto Bioscience which is now part of BD Biosciences was able to effectively build a biosensor selective for cAMP that signals through calcium or membrane potential responsive dyes.
